Selective enrichment method for glycopeptide is challenging and key problems to glycoproteomics, which is vital to explore the pathogenesis of disease, discover disease biomarkers and develop new drugs. Lectin shows specialty to glycoproteins, which is determined by special steric structure, multiple interaction sites and hydrogen bound between peptides and glycans. Inspired from specific affinity of lectin to glycoproteins, functional units including oligopeptide, oligosaccharides and their combination and main polymer chains will be grafted onto the silica microspheres with surface-initiated polymerization method. The interaction machanism between smart polymer materials and glycopeptides will be explored. Based on these, the seletive enrichment method for glycopeptides will be established and validated with serum samples. Compared with currently used materials modified with small molecules, introducing smart polymer materials into the field of glycopeptide enrichment and separation will benefit the separation materials several advantages, including higher selectivity, higher adsorption capacity and easier controlloed operating conditions. These advantages are atributed to the multiple interaction sites, affinity interaction, steric compatibility between materials and glycans, and multiple respondiness provided by smart polymer materials. we believe that introducing smart polymer materials into the field of separation will provides an original strategy for the glycopeptide enrichment.
高选择性糖肽富集方法的发展是糖蛋白组学中的难点问题,对研究疾病发生机理、发现疾病标记物及开发新药具有重要意义。受启示于凝集素对糖蛋白的特异性识别取决于凝集素特定的几何结构、高度多价性和肽与糖之间的氢键作用,本项目拟在多响应聚合物主链中引入寡肽、寡糖或二者组合的功能单元,制备用于高选择性糖肽富集的智能聚合物材料,探讨糖肽与材料之间的作用机理;建立材料对糖肽富集方法,并在血清样品中进行验证。相比于现有的小分子修饰糖肽富集材料,除了小分子与糖肽的作用外,研制的智能聚合物材料还具有多价性、与糖链匹配的空间和多响应性等优势;基于智能聚合物材料的糖肽富集方法选择性更高选择性、吸附量更大和可控性更好。智能聚合物材料的引入为糖肽的富集和分离提供了一种新方法、新思路。
