Embryo implantation is regulated strictly by microenvironment. This proposal was designed to investigate the regulation role of insulin-like growth factor(IGF) autocrine/paracrine system during embryo implantation. We established the pregnant model of rat during 3-10 day of pregnancy ,and the rat in estrus was used as control. The serum progesterone levels in rats and women at 5~7 weeks of gestation were measured by radioimmunoassay.The expressions of IGF-I, IGF-IR, IGFBP-1,IGFBP-2,IGFBP-3 and progesterone receptor (PR) in endometrium/ decidua and trophoblast in rats and women at 5~7 weeks of gestation were detected by histoimmunochemistry and quantified by computer image analysis. And we also examined the mitogenic activity of IGF-I, IGF-II and progesterone by [3H]thymidine incorporation and the effect of progesterone on the expression of mRNAs encoding IGF system in cultured human decidual cells and trophoblast cells by reverse transcriptase polymerase chain reaction (RT-PCR). P and PR showed significant correlation with implantation window. The expression of IGF-IR in endometrium/decidua was decreased significantly by progesterone,while the expressions of IGFBP-1,IGFBP-2 and IGFBP-3 were increased. Furthermore,IGF-I, IGF-II and progesterone stimulated proliferation of human decidual cells and trophoblast cells. The results demonstrated that progesterone has some important roles in embryo implantation by increasing the expressions of IGFBP-1,IGFBP-2 and IGFBP-3 while decreasing the expression of IGF-IR, and which will satisfy the need of endometrial decidulization and embryo growth.In our study, it is suggested that progesterone may be the promotor of implantation window,its function is partly mediated by IGF system.
胚泡着床受微环境严格调控,通过建立着床过程的动物模型和人体研究,对着床前子宫内膜和着床后早服蜕膜和绒毛细胞及细胞,用免疫组化、计算机图像分析和分子杂交技术系统检测胰岛素样生长因子(IGFs),IGFsmRNA,孕激素受体的表达及细胞增殖活性。探讨孕激素通过IGFs介导,调节子宫内膜及早孕蜕膜和绒毛的发育,影响胚泡着床及早期妊娠建立的机理。
