Essence can change into blood, while blood can change into essence too (i.e. essence and blood sharing the same origin), it’s the fundamental theory of traditional Chinese medicine. It is recently found that PLCγ plays an important role in the process of microcirculation of the testis ,it can participate in the formation of NO in the testicular interstitium,and it can regulate the the testicular microcirculation. Previously, we demonstrated that Yangjing capsule can improve testicular microcirculation and promote testicular spermatogenic function by up-regulation of VEGF and VEGFR, NO and NOS.Therefore, we hypothesized that the Yangjing capsule could improve the expression of AKT/eNOS through PLCγ, promote the synthesis of NO, improve the testicular microcirculation, and promote the spermatogenic function.This study intends to use adult male mice, cell experiments, and small interfering RNA technology to further study the effect of kidney reinforcing drug on the expression , location and promotor of PLCγ.Then it can be identified that whether the mechanism of improving testicular microcirculation to promote testicular spermatogenesis through the role of PLCγ.The molecular mechanisms that tonify kidney and nourish blood improve the microcirculation of male reproductive function targets would be clarified,the study would contribute to elaborate the scientific connotation of “essence and blood sharing the same origin”,Therefore, it is of great theoretical significance and clinical value to guide the clinical application of traditional Chinese medicine in the treatment of male infertility caused by oligospermia.

精能化血,血亦能化精,即“精血同源”是中医的基本理论。近年发现磷脂酶Cγ(phospholipase C, PLCγ),在改善睾丸微循环,促进睾丸生精功能的过程中发挥重要作用,它能够参与睾丸间质内NO的生成,调节睾丸微循环。前期研究表明,补肾养血中药—养精胶囊能够通过上调VEGF及VEGFR,NO及NOS改善睾丸微循环,促进睾丸生精功能。所以课题组推测养精胶囊能可能是通过PLCγ上调AKT/eNOS的表达,促进NO的合成,改善睾丸微循环,促进生精功能。本研究拟利用成年雄性小鼠、细胞实验及小干扰RNA技术,进一步研究补肾养血中药对PLCγ的表达、定位和上游启动子的影响,明确养精胶囊通过PLCγ改善睾丸微循环促进睾丸生精功能的作用机制。这对于阐明补肾养血法改善微循环男性生殖功能的作用靶点,揭示“精血同源”的科学内涵,进而指导临床运用中药治疗少弱精症导致的男性不育,具有重要的理论意义和临床价值。

项目摘要

精能化血,血亦能化精,即“精血同源”是中医的基本理论。近年发现磷脂酶Cγ(phospholipase C, PLCγ),在改善睾丸微循环,促进睾丸生精功能的过程中发挥重要作用,它能够参与睾丸间质内NO的生成,调节睾丸微循环。前期研究表明,补肾养血中药—养精胶囊能够改善睾丸微循环,促进睾丸生精功能。本项目基于前期研究和精血同源理论的支持,利用奥硝唑制造的大鼠生精损伤和睾丸微循环障碍模型,细胞实验及小干扰RNA技术,进一步证明了养精胶囊可以通过 PLCγ促进 AKT、eNOS合成及提高NO含量,进而改善睾丸微循环促进睾丸生精功能。这对于阐明补肾养血法改善微循环男性生殖功能的作用靶点,揭示“精血同源”的科学内涵,进而指导临床运用中药治疗少弱精症导致的男性不育,具有重要的理论意义和临床价值。
