Through EST which had prominent decreasing expression in NPC, a novel tumor suppressor candidate NPCDR1 was cloned by 5',3'-RACE. By transfection of NPCDR1 gene into CNE2 with NPCDR1 down-regulation, the growth rate of transfected cells slowed down. The doubling time was prolonged. The colony formation rate was significantly decreased (p<0.01). The growth rate of nude mice tumor was inhibited, and it was identified as high-differentiated squamous carcinoma by pathological method. The results are important clues and bases for clarifing the molecular mechanism of carcinogenesis and development and studing the prevention and treatment of NPC.
本项目采用新近建立的充分利用生物信息数据库资源进行基因分析、且能鉴定和克隆稀有拷贝基因的全基因组基因研究策略——基因鉴定集成法(IPGI),分别研究鼻咽癌和鼻咽正常上皮的基因组表达状况,克隆和分离差异表达的与癌变相关的新基因。这是了解和揭示鼻咽癌患病易感性的重要关键,对阐明鼻咽癌发生的分子生物学机制具有重要理论价值。
