Previously, we observed clinically that resistant human papilloma virus infected diseases could be cleared by local hyperthermia.The number of FOXP3 was significantly increased in the resistive patients than nonresistant patients. And in the regressing lesions, we found that the FOXP3 is decreased than in the no-regressing lesions. Indicating that CD4+CD25+FOXP3+Tregs may play a role in the anti-HPV immunity responses.However, the regulation of cellular immune responses especially the CD4+CD25+FOXP3+Tregs in genital warts during local hyperthermia treatments is unclear to date.This study could contribute to further understanding of the relations of Tregs, CCR4-CCL17 and langerhans cell in anti-HPV immunity responses, and bring further understandings into the mechanism of anti-viral immunity induced by local hyperthermia. And improving the cure rate for the local hyperthermia treatment of HPV related diseases.
前期临床研究发现,局部温热治疗一些难治性人乳头瘤病毒感染性皮肤病取得良好临床疗效,且发现在一些难治性的尖锐湿疣皮损局部Foxp3与非难治性尖锐湿疣相比明显增多,提示 CD4+CD25+FOXP3+Tregs的聚集可能是HPV局部免疫逃逸的机制之一。且在温热干预后呈退行性改变的HPV感染组织中FOXP3明显减少,提示温热可能通过影响局部CD4+CD25+FOXP3+Tregs而激发了机体针对HPV的有效免疫。本研究以温热干预的疣体组织为研究对象,研究温热对皮肤内朗格汉斯细胞、CCR4-CCL17等相关信号传导通路,进而影响了CD4+CD25+FOXP3+Tregs游走、分化、归巢以及免疫应答及调节作用,寻找关键分子干预靶点的可能性,深入认识温热诱导抗病毒免疫机制,提高局部温热疗法治疗HPV感染相关疾病的临床治愈率。
