The discovery of natural nuclear factor erythroid-2 related factor 2 (Nrf2) activators has been proved to be an effective way in developing therapeutic drugs on diabetic nephropathy (DN). The water extract of Tadehagi triquetrum was confirmed to possess obvious protective effects on DN in our previous study. Further pharmacological investigations have been successfully carried out by which the active fraction was determined. Tentatively chemical investigation indicated that the active fraction is rich in phenylpropanoid glycosides (PPGs). Furthermore, PPGs obtained showed significant anti-oxidative stress activities and the mechanism was found to be closely associated with activating the Nrf2. Based on the previous work, our research group aim to systematically investigate PPGs with therapeutic effects and molecular mechanism on DN in the active fraction of Tadehagi triquetrum. By comprehensive application of natural medicinal chemistry, pharmacology, molecular biology and other related methods, PPGS will be isolated and structurally elucidated. Following, the anti-oxidative stress activities and activation of Nrf2 will be thoroughly screened in vitro. The compounds with novel structures, better biological activities will be selected and prepared for large scale. The pretective effects on DN of the selected PPGs will be tested using animal model. Moreover, the molecular mechanism will be systematically investigated. In this way, our findings will find new therapeutic agents with novel chemical structures, obvious effects and confirmed mechanism on DN. Collectively, the completion of this project will establish solid scientific foundation, facilitating not only research and develop this Li medicine but also find new protective drugs on DN with independent intellectual properties.
核因子E2相关因子2(Nrf2)天然激活剂的发现是糖尿病肾病(DN)保护药物研发的有效途径。课题组前期发现黎药-葫芦茶水提物的DN保护作用显著,进一步确定其有效部位。深入研究发现该有效部位富含结构新颖、活性显著的苯丙素苷类成分(PPGs),且此类成分可能通过激活Nrf2发挥DN保护作用,具有重要的科学研究价值。本项目拟在前期工作基础上,综合运用天然药物化学、药理学、分子生物学等研究方法,实现有效部位中新颖PPGs的分离、制备与结构鉴定;体外评价所得成分的抗氧化应激及Nrf2激活作用;遴选结构新颖、活性显著的化合物,足量制备;完成遴选成分体内DN保护作用的系统评估,并阐释活性成分通过激活Nrf2发挥功效的分子机制。通过本项目的实施,从葫芦茶PPGs中发现结构新颖、活性显著先导化合物,阐明其药理作用的分子机制,为葫芦茶的深入研究与开发提供科学依据,为形成具有自主知识产权的DN保护药物奠定基础。
黎药葫芦茶在海南有着广泛的应用,对于糖脂代谢调控效果优良,课题组前期发现葫芦茶有效部位可显著改善糖尿病肾病,而且有效部位中富含新颖的苯丙素苷类化合物,且这些化合物发挥功效与抑制Nrf2密切相关。本项目完成了有效部位中化学成分的研究,从中分离得到36个化合物,12个为苯丙素苷类化合物,10个为新化合物,结构非常新颖;完成对有效部位中所得苯丙素苷类化合物降糖作用进行评估,发现苯丙素苷类化合物及部分黄酮苷类化合物可促HepG2细胞葡萄糖消耗,并促进肌细胞C2C12对葡萄糖的吸收,具有明显的降糖作用,特别是课题组发现的Tadehaginoside D降糖作用甚至优于同等剂量的罗格列酮,并发现这些苯丙素苷类化合物具有明显的改善动脉粥样硬化作用完成对活性显著的苯丙素苷类化合物调控Nrf2进行评价,发现部分化合物可以显著激活Nrf2及其下游靶蛋白和基因的表达。葫芦茶苷是葫芦茶中含量最高、活性较好的新颖苯丙素苷类化合物,为了评估其成药性,完成了药代动力学、组织分布、体内代谢活性产物等研究,明确了葫芦茶苷的半衰期、主要分布器官,特别是发现葫芦茶苷的活性代谢产物对羟基桂皮酸。完成对有效部位改善胰岛素抵抗的作用研究,发现有效部位可显著降低ob/ob小鼠的血糖(空腹血糖、口服葡萄糖耐量)和血脂水平(TC、TG、LDL-c),并对有效部位中8个主要成分进行活性测试,发现2个苯丙素苷类化合物活性显著。在完成这些工作的同时,课题组对葫芦茶中所含的化合物丁香树脂酚-4-O-葡萄糖苷降脂和降糖作用进行研究,并探索其作用机制。目前,正在开展有效部位对糖尿病肾病作用和葫芦茶苷的人工合成。本项目基本完成既定的研究内容和目标,发表SCI收录论文5篇:Acta Pharm Sin B,Journal of Natural Products,Natural Product Research,Tropical Journal of Pharmaceutical Research,Chem Pharm Bull,投稿SCI论文1篇(Biomedical Chromatography),发表中文期刊3篇,授权国家发明专利1项,指导舒佳慧荣获第四届全国大学生基础医学创新论坛暨实验设计大赛一等奖,获2018年度海南省自然科学奖三等奖一项(第一完成人,2018-Z-3-10-R01)。
