Whitefly transmitted Geminivirus(WTGs) belongs to Begomovirus, Geminiviride. The virues of this genus do much damage to many crops in the tropics and subtropics area, moreover, it is easy to produce new strains because of the DNA recombination, which would cause serious epidemic damage. Yunnan Province is situated in stereoscopic climate zone covering area of low latitude and various altitude, most of the area are in the tropics and subtropics climate zone. The virus disease caused by WTGs were found widely in tobacco, aromatic tobacco, and white burley, furthermore, the incidence of disease was over 30% in some area. Therefore, it is a kind of dangerous virus disease. Supported by National Natural Science Foundation of China in 2000, we carried out the study of Yunnan whitefly transmitted Geminivirus on biology and molecular biology, cooperating with the Biotechnology Institute of Zhejiang University and Scottish Crop Research Institute. By the detection of indicative plant, triple antibody sandwich enzyme linked immunnorsorbent assay, PCR and sequence analysis of the WTGs antigenic region and conservation region of CP gene, PCR and sequence analysis of the genome of DNA-A, DNAβ, we found that WTGs distributed widely in the area of tropics and subtropics below altitude 1500 meters infecting tobacco, tomato, pumpkin, capsicum and etc. It caused the symptom of leaf curl, stem curl, enation at tobacco and tomato the symptom of mosaic, vein enation at pumpkin and the symptom of ferny leaf, crinkle at tobacco and tomato, the symptom of mosaic, vein enation at pumpkin and the symptom of ferny leaf, crinkle at capsicum. The weeds in the field such such as Ageratum conyzoides Linn, Malvastrum coromandelianum, Siegesbeckia pubescens Mak. are the important interim hosts. The disease was more serous in the area altitude below 1000 meters while no disease was found in the area altitude above 1500 meters. Two distinct species of Begomovirus respectively named Tobacco leaf curl Yunnan virus(TLC Yn V) and Tobacco curly top virus(TCTV) were isolated from aromatic tobacco in Yunnan Province. The other virus isolates belong to Tobacco leaf curl virus(TLCV) and Tomato leaf curl irus(ToL CV). Two new interim host were found, with typical symptom of vein yellow, which could be indicative plant. Aew accompanied DNA β was identified in the isolate of Tobacco leaf curl virus from tobacco. The molecule was affirmed to be contained in the virus particles by the detection of IC-PCR, and transmitted by whitefly. The distribution of WTGs in Yunnan Province related to the climatic types in which host and intermedia existing but not the host variety and the distribution of intermedia themselves.
对粉虱传双生病毒(WTG)的单克隆抗体,运用ELISA和电镜技术对云南省主要作物上WTG进行检测,比较其生物学差异,在此基础上利用简并引物PCR扩增有关分离物基因组的共同区和外壳蛋白基因,进行克隆和序列测定,并进而克隆全基因组,分析基因结构。可为了解云南省WTG的发生分布及防治打下基础。.
