黄连素对肠道平滑肌细胞膜钙和钾通道电流的影响

Effects of berberine on IKV , IKCa , ICa-L of guinea pig colonic smooth muscle cells were studied by patch clamp technique. Effects of berberine on [Ca2+]i of guinea pig colonic smooth muscle cells were studied by Fura 2-AM fluorescence technique. Effects of berberine on guinea pig colonic smooth muscle contraction were also studied.It's showed that 10μM ~100μM berberine can inhibit IKV , IKCa , ICa-L of guinea pig colonic smooth muscle cells significantly, berberine can inhibit kcl,Ach, CPA induced [Ca2+]i increase.Berberine can significantly inhibit contraction of longitude colonic smooth muscle and circuit smooth muscle on its extent ，but no significant inhibition effect on its frequency.All above clarified the mechanism of berberine cure motility diarrhea,it's significative for further use of berberine..keywords:Berberine; Patch clamp; Colonic smooth muscle cells

本项目拟使用膜片钳技术研究黄连素对豚鼠结肠平滑肌细胞膜钙和钾通道电流以及对乙酰胆碱效应的影响,探讨黄连素抑制肠道运动与肠道平滑肌细胞膜钙、钾离子通道的关系,从而阐明黄连素治疗运动性腹泄的机制,为进一步开发黄连素的临床药用价值奠定理论基础.
